Abstract:

A new, simple, rapid, selective, precise, and accurate isocratic reverse-phase high-performance liquid Chromatography assay method has been developed for simultaneous estimation of Piracetam and Mecobalamin tablet formulations. The separation was achieved by using column Phenomenex Luna C18, 250x4.6mm, 5μm, in mobile phase consisted of pH 6.0 phosphate buffer and Acetonitrile and methanol in the ratio of 40:50:10 v/v/v. The flow rate was 1.0 mL/min, column oven temperature 25° C, sample cooler temperature 25°C and the injection volume was 20μL, and detection was performed at 215 nm using a photodiode array detector (PDA), Run time 7minutes. The retention time of Piracetam and Mecobalamin was noted to be 2.218 minutes and 3.68 minutes, respectively, indicative of rather shorter analysis time. The method was validated as per ICH guidelines. The proposed method was found to be accurate, reproducible, and consistent.
